| Abstract | A Gram-positive strain of Alkalihalobacillus clausii ABF28, found in environments contaminated with Cr(VI), was identified through molecular imaging and de novo genomic profiling. To evaluate the remediation efficiency of this bacterial strain, it was cultivated in a Novel Column Bioreactor designed to achieve a higher retention time. The minimum inhibitory concentration (MIC) for the indigenous strain was determined to be 2500 mg/l. X-ray photoelectron spectroscopy (XPS) confirmed that the bacterial strain successfully reduced Cr(VI) to a +3 oxidation state. The strain was cultivated in both free and immobilized forms and subsequently analysed using scanning electron microscopy (SEM), XPS, and Fourier-transform infrared spectroscopy (FTIR). A similar finding was observed in the elemental mapping conducted via energy-dispersive X-ray spectroscopy (EDX).Molecular characterization through whole genome sequencing identified the genes responsible for the reduction of chromium (VI) to chromium (III) via the chromate reductase enzyme in this study. Given its notable chromium removal capability and strong resilience, this bacterial isolate offers a promising solution for the biotechnological remediation of chromium and other heavy metals from environments contaminated with trace metals. |
